Light-Triggered Control of Thiol–Maleimide Click Chemistry
Thiol–maleimide click chemistry has been extensively studied as an efficient and selective route for covalent coupling between thiol and maleimide functionalities. Despite its advantages, precise spatial and temporal control over the initiation of this reaction remains a key limitation. Photo-switchable systems (e.g., spiropyrans) provide a promising strategy for light-controlled activation, but their limited stability in aqueous environments constrains their applicability in biological contexts. Combining in computational simulations with targeted experimental synthesis and validation, we aim to design and realize water-stable, light-responsive molecular switches suitable for biological use.
